ABSTRACT:
A multistep synthesis was presented for the prepn. of 4,5-diaminoshikimic acid derivs. I [R1, R2 = alkyl; R3, R4 = H, acyl, etc., except R3 = R4 = H], which are potent inhibitors of viral neuraminidase, via Diels-Alder cycloaddn. of furan and Et acrylate and lipase mediated hydrolysis. Thus, Tamiflu I (R1 = CHEt2, R2 = Et, R3 = COMe, R4 = H) was prepd. via a multistep synthetic sequence starting by Diels-Alder cycloaddn. of furan and Et acrylate to form ester (.+-.)-II in 80% yield by followed by Chirazyme L-2 mediated resoln. to yield the of the desired (1S,2R,4S)-II.
