ABSTRACT:
An enzymic RNA mol. is presented which specifically cleaves mRNA encoded by an mdr-1 gene, or assocd. with development or maintenance of chronic myelogenous leukemia, promyelocytic leukemia, Burkitt's lymphoma or acute lymphocytic leukemia, follicular lymphoma, B-cell acute lymphocytic leukemia, breast cancer, colon carcinoma, neuroblastoma, and lung cancer. These cancers are treated by administering the enzymic RNA. The enzymic RNA cleaves target mRNA having sequences presented. A hammerhead ribozyme was synthesized and its stability was tested in Vero and HeLa cell fractions. Liposomes contg. 15 ribozymes were prepd. and incubated with cells.
