The total synthesis of ecteinascidin 743 (I), an extremely potent antitumor agent, has been accomplished. The synthesis features Ugi's four component coupling reaction, intramol. Heck reaction, phenol-aldehyde cyclization, and acid-induced intramol. sulfide formation.
