ABSTRACT:
A reshaped human monoclonal antibody (MAb) to human interleukin-6 receptor (IL-6R) is provided. The light (L) chain of the MAb consists of (1) a variable (V) region that is comprised of a framework (FR) of a human L chain and the complementarity detg. region (CDR) of the L chain of a mouse MAb and (2) a const. (C) region of human L chain. The heavy (H) chain of the MAb is also similarly humanized. The chimeric MAb is useful as a therapeutic agent. Humanizing MAb PM-1 and AUK12-20 using the FR of the L chain of human antibody REI and the FR of te H chain of NEW or HAX by expression of the chimeric genes in animal cells is described.
