ABSTRACT:
Title compds. [I:R1 = H, (un)branched C1-6 alkyl or alkoxy, OH, SH, C3-6 cycloalkyl, morpholinyl, piperidinyl, etc.; R3 = (un)substituted alkyl, alkenyl, etc.; X1 = direct bond, CH2, O, S, NR; R = H, alkyl; Y = Ph, thienyl, benzothienyl pyridyl, quinolyl, pyrazinolyl, pyrimidinyl, etc.; Z = substituted amino, heterocyclo, (un)substituted alkoxy, heterocyclo, etc.], which have ACTH-releasing factor (CRF) antagonistic activity (no data) and are useful in the treatment of illnesses induced by CRF [e.g., stress- and anxiety-related disorders (no data), etc.], are prepd. Thus, 2-[5-methyl-3-methylthio-1-(2,4,6-trichlorophinyl)pyrazol-4-yl]methyl-1,2,3, 4-tetrahydroisoquinoline hydrochloride, m.p. 205-207.degree. was prepd. in 53% overall yield from Et acetoacetate in 3 steps.
