ABSTRACT:
Pipet 2 ml. of sugar soln. contg. 10-70 .gamma. of sugar into a colorimetric tube. Add 0.05 ml. 80% phenol and 5 ml. concd. H2SO4. Mix thoroughly, allow to stand for 10 min., and then place the tubes in a H2O bath at 25-30.degree. for 10-20 min. Measure the absorbance of the soln. at 490 m.mu. for hexoses and 480 m.mu. for pentoses and uronic acids. Det. the amt. of sugar from standard curves prepd. for each particular sugar. Sep. sugar mixts. by paper chromatography. Cut the paper into sections and elute each with 20 ml. H2O. Filter the eluate through glass wool and det. the sugar concn. as given above.
