Abstract:
The present invention pertains to a process for the preparation of 1,7-dichloro-4-methoxy-isoquinoline. For example, 5-chloro-2-methylbenzoic acid was treated with oxalyl chloride and then with NH4OH for 5-chloro-2-methylbenzamide, which was then reacted with 1,1-dimethoxy-N,N-dimethyl-methanamine and further treated with potassium-t-amylate for cyclization product, 7-chloro-1(2H)-isoquinolone. The intermediate obtained above was reacted with methanol in the presence of methanesulfonic acid a hypervalent iodine oxidizing agent, e.g., Ph iodine diacetate, to afford 7-chloro-4-methoxy-1(2H)-isoquinolinone, which was then treated with a chlorinating agent, e.g., phosphoryl chloride, to give 1,7-dichloro-4-methoxy-isoquinoline as the final product.
