CAPLUS COPYRIGHT 2012 ACS

Patent Number: WO 2012064642
Title: Preparation of spirocyclic piperidine derivatives as fatty acid synthase inhibitors for treatment of cancer
Inventor(s): Adams, Nicholas D.; Chaudhari, Amita M.; Kiesow, Terence John; Parrish, Cynthia Ann; Reif, Alexander Joseph; Ridgers, Lance Howard; Schmidt, Stanley J.; Wiggall, Kenneth
Patent Assignee(s): GlaxoSmithkline LLC, USA
Source: PCT Int. Appl., 221pp. CODEN: PIXXD2
Language: English

Abstract: The title compd I [R3 = (un)substituted C1-6 alkyl, (un)substituted C3-7 cycloalkyl, (un)substituted C4-6 heterocycloalkyl; R4 = oxo, halo or C1-6 alkyl; Cy = (un)substituted Ph, (un)substituted pyridinyl or (un)substituted 5- or 6-membered heteroaryl; R1 = (un)substituted Ph, (un)sustituted 5- or 6-membered heteroaryl, (un)substituted naphthyl, etc.; R7 = independently H, C1-3 alkyl, halo, etc.; X = CH2, NR6 or O, wherein R6 = H, C1-4 alkyl, C3-7 cycloalkyl, etc.; n = 0, 1, 2, 3 or 4; or a pharmaceutically acceptable salt thereof] was prepared. Compd II was prepd in a multi-step synthesis. Compd II displayed IC50 of 6 nM against FAS. Compd I was claimed useful for treatment of cancer.

Please note:  Titles and abstracts for patent records in CAplus are routinely enhanced to better describe the claimed invention. In this example, CAS enhanced both the abstract and title using additional information found in the patent. The original title of the PCT publication is Fatty acid synthase inhibitors. For the CAplus record, this title was enhanced to read Preparation of spirocyclic piperidine derivatives as fatty acid synthase inhibitors for treatment of cancer.
